摘要
目的探讨葛根素对急性视网膜缺血再灌注兔眼模型视网膜组织中游离谷氨酸(gluta-mate)浓度的影响。方法新西兰白兔20只随机分为对照组和实验组,各组右眼应用前房灌注加压法使前房内压力升高至120 mm Hg维持60分钟后再灌注,建立视网膜缺血再灌注模型,左眼为空白对照眼。实验组在前房灌注前2小时视网膜组织腔内注射葛根素溶液,对照组给予等量生理盐水视网膜组织腔内注射。再灌注24小时摘除眼球,采集视网膜组织,研磨及蛋白沉淀后用日立L-8800氨基酸自动分析仪检测游离氨基酸浓度。结果对照组兔右眼视网膜组织中游离谷氨酸浓度较左眼升高,差异有显著性意义(t=9.518,P<0.001)。实验组兔右眼视网膜组织中游离谷氨酸浓度较左眼升高,差异有显著性意义(t=2.519,P<0.05);但较对照组浓度低,差异有显著性(t=6.575,P<0.05)。结论兔视网膜缺血再灌注可致视网膜组织中游离谷氨酸浓度升高,是造成视网膜组织损伤的可能机制之一;而葛根素可以通过阻止谷氨酸浓度升高,从而保护了视网膜组织。
OBJECTIVE To study and discuss the effect of puerarin on Glutamate (Glu) concentration in rabbit retina in retinal ischemic - reperfusion injury. METHODS Twenty rabbit were divided into two group, the control group and the treatment group. Of all animals, retinal ischemia- reperfusion models were induced in the right eyes by increasing intraocular pressure to 120mm Hg for 60 minutes; the left eyes were sham operated as the control eyes.The treatment group was intravitreal injected puerarin before ischemia. The retina was collected separately on 24 hours after reperfusion. Glu levels were analyzed by HITACHI L- 8800 Amino Acid Analyzer. RESULTS In the control group, the Glu concentration in right eye retinas was significantly higher compared with the left eye ( t = 9.518, P 〈 0.001) . While the retinal Glu level was increased significantly compared with the left eye (t = 2.519, P 〈 0.05), and have statistical difference compared with the right eyes of control group ( t = 6. 575, P 〈 0.05) . CONCLUSIONS Retinal ischemic- reperfusion injury can result in retinal Glu concentration, and may be one of the mechanisms of retinal ischemic - reperfusion injury; while puerarin can stop the over release of Glu, and prevent the retina ischemic - reperfusion injury.
