以啤酒废酵母为原料,采用酶-碱法提取酵母碱不溶性葡聚糖,确定了酵母碱不溶性葡聚糖的制备条件:50g酵母泥加入200mL浓度为20mg/L木瓜蛋白酶溶液,55℃处理36h,离心所得沉淀物,以1∶5固液比(w/v)加入1.0mol/LNaOH溶液,45℃处理3h。此条件下多糖得率为10.1%,多糖纯度达到92.6%。红外光谱分析显示产品含有β-(1-3)键葡聚糖。
The alkali-insoluble glucan was isolated by enzyme-alkali method from waste beer yeast. The relative preparation conditions were as follows: 50 g yeast slurry added in 200 mL papain solution (its concentration as 20 mg/L), then after 36 h treatment at 55 ℃, the precipitate obtained through centrifugation, then added in 1.0 mol/L NaOH solution (solids: solution =1:5), and then 3 h treatement at 45 ℃. Alkalic-insoluble glucan yield was 10.1% and the content of glucan was as high as 92.6 %. Infrared spectra analysis showed that the product contained β-(1-3) glucan.
