摘要
目的:研究肝卵圆细胞增殖模型大鼠Notch1,Jag-ged1mRNA表达的变化及苦参碱对其的影响.方法:雄性SD大鼠48只按体重随机分为4组:模型组(A组);苦参碱小剂量(B组);苦参碱组大剂量(C组);D组即正常对照组.用免疫组化方法观察造血干细胞标记(Thy-1)表达的变化、半定量RT-PCR方法观察肝组织中Notch1,Jagged1mRNA表达的变化.结果:与D组比较,A组Thy-1蛋白,Notch1mRNA,Jag-ged1mRNA表达显著升高(P<0.01),与A组比较,C组Thy-1蛋白,Notch1mRNA,Jagged1mRNA表达显著下调(P<0.01).结论:苦参碱可有效的调节Notch信号通路、下调Thy-1蛋白表达,可能是其抑制卵圆细胞过度增殖、诱导卵圆细胞向肝细胞方向定向分化的机制之一.
AIM: To study the expressions of Notch1, Jagged1 mRNA in hepatic oval cell proliferation model rats and the effect of matrine on them. METHODS: Forty-eight male SD rats were randomly separated into 4 groups according to body mass: model group( group A) ,low-dose matrine group( group B, 5 mg/100g), high-dose matrine group (group C, 25 rag/100 g) , control group (group D). The expression of Thy-1 protein was detected by enzyrnehistochemisty, and the expressions of Notch1, Jagged1 mRNA were detected by semi-quantitative RT-PCR method. RESULTS: Compared with group D , the expressions of Thy-1 protein, Notch1 mRNA, Jagged1 mRNA in group A were singnifieantly increased (P 〈 0.01 ). Compared with group A, the expressions of Thy-1 protein, Notch1 mRNA,Jagged1 mRNA in group C were singnificantly increased ( P 〈 0. 01 ). CONCLUSION: Matrine could regulate Notch signaling, decrease the expression of Thy-1 protein ,which might be one of the mechanisms of inhibiting the proliferation of hepatic oval cells and inducing rat hepatic oval cells to differentiate into mature hepatocytes.
