褪黑素对Aβ_(25-35)诱发PC12细胞损伤的保护作用 被引量:1

摘要 目的研究褪黑素(MT)对β淀粉样蛋白片段(Aβ25-35)诱发PC12细胞损伤的神经保护作用。方法将培养的PC12细胞分为三组:正常对照组,Aβ损伤组和MT保护组。Aβ损伤组用Aβ25-35处理细胞,MT保护组在用Aβ25-35处理前2 h加入褪黑素。使用细胞形态学方法、LDH法、MTT法及W estern印迹法观察MT的神经保护作用。结果形态学观察发现MT保护组细胞数显著比Aβ损伤组多,受损变圆的细胞数较少。MT保护组LDH活性显著低于损伤组(P<0.01);OD值显著高于损伤组(P<0.01)。W estern印迹结果表明Aβ25-35损伤组中Bcl-2的表达比正常对照组低;MT保护组Bcl-2的表达比Aβ25-35损伤组升高。结论褪黑素对Aβ25-35诱发PC12细胞损伤有保护作用。 Objective To study the protective effects of melatonin (MT) on injury of pheochromocytoma (PC12) cells induced by amyloid 13 (A[325-35) protein. Methods Cultured PC12 cells were divided into normal control group, Aβ group (treated by Ab25-35) and MT group (added with MT 2 h before treated by Aβ25-35). Morphological observation, MTr assays, LDH assays and were used to observe the protective effects of MT. The level of Bcl-2 in PC12 cells was detected by Western blot method. Results Morphological observation showed cell number was more and less rounded injured cells in MT group than those in Aβ group. LDH activity was less ( P 〈 0. 01 ), OD was significantly higher ( P 〈 0.01 ) in MT group than those in Aβ group. Western blot showed Bcl-2 expression in Aβ group was higher than that in control group, but was lower than that in MT group. Conclusions MT could play the protective role in PC12 ceils injury induced by Aβ25-35.
