ATRA诱导HPV16型亚基因永生化人宫颈上皮细胞分化的实验研究 被引量:1

摘要 目的:研究ATRA对HPV16型亚基因(E6、E7)永生化人宫颈上皮细胞(H8细胞)的诱导分化作用及机制。方法:H8细胞经ATRA处理后,用MTT法测定ATRA的抗增殖效应,光镜、电镜观察细胞形态变化,流式细胞术(FCM)检测细胞周期,免疫组化SABC法检测增殖细胞核抗原Ki67表达水平的变化,PCR-ELISA法检测端粒酶活性变化,荧光免疫细胞化学检测HPV和E6蛋白表达变化。结果:ATRA使H8细胞增殖抑制;细胞形态朝良性方向分化;细胞堆积于G1期,S期细胞减少;细胞核内Ki67的表达水平下降;端粒酶活性下降。结论:ATRA对H8细胞有明显诱导分化作用,这种作用可能是通过抑制增殖,阻滞细胞周期,降低端粒酶活性来实现的。 Objective:To study the differentiation-inducing effects of all-trans-retinoic acid (ATRA) on HPV16 subgenes-immortalized human endocervical cells (H8 cell) and the mechanism. Methods:After H8 cells were treated with ATRA, the aiti-proliferation of ATRA on them was tested by MTT assay, morphological changes observed by LM and TEM, cell cycle analyzed by FCM, expression of Ki67 tested by immunocytochemistry, the activity of telomerase tested by PCR-ELISA, and expressions of HPV and E6 tested by fluorescence immunocytochemistry. Results:ATRA could inhibit proliferation of H8 cells and induce their benign morphedifferentiation. According to FCM, H8 cells accumulated in G1 phase after being treated with ATRA, and expression of Ki67 and activity of telomerase reduced. Conclusion:ATRA could induce the differentiation of H8 cell line, which might be achieved by inhibiting proliferation, blocking cell cycle, and decreasing activity of telomerase.
