摘要
本文采用快速印迹杂交(quickblot)和延续转录试验(run-oftranscriptionassay)两种特异mRNA检测技术,在经β-萘黄酮(β-NF)诱导的人羊膜上皮细胞FL系中,观察了茶多酚对人羊膜上皮细胞FL系CYP1A1mRNA水平和转录的影响,结果表明,细胞中CYP1A1mRNA的含量和细胞核CYP1A1转录水平均比空白对照组增加,茶多酚可明显抑制其基础表达及β-NF的诱导作用。因此,我们认为茶多酚预防肿瘤的作用还可能与它影响细胞CYP1A1基因表达,从而影响CYP1A1同功酶活性有关。
By means of quick blot and run-off transcription assay,the effect of green tea polyphenols (GTPP) on CYP1A1 mRNA levels and transcription in human amnion cells (FL cells) induced by β naphthoflavone (β NF) was studied.The results showed that β NF(2×10 -5 mol/ml) could increase CYP1A1 mRNA levels and enhance the CYP 1A1 transcription in FL cells,both of the constitutive and induced CYP 1A1 transcription could be inhibited by GTPP at the concentration of 50 and/or 100μg/ml.
