摘要
目的:探讨hTERTmRNA及hTERT蛋白在口腔颌面部肉瘤组织中表达的生物学意义。方法:应用原位杂交和免疫组化技术检测37例口腔颌面部肉瘤和14例良性间叶组织肿瘤中hTERTmRNA和hTERT蛋白的表达,探讨口腔肉瘤组织中hTERT表达的生物学意义。结果:肉瘤中hTERTmRNA的阳性表达率明显高于良性肿瘤(62.2%/14.3%,P<0.05)。在低度、中度和高度恶性肉瘤组织中,hTERTmRNA阳性表达率分别为41.67%、50.00%和86.67%,统计分析显示低度恶性与高度恶性组间差异有统计学意义(P<0.05)。复发肉瘤中hTERTmRNA的阳性表达率明显高于原发肉瘤(P<0.05)。在发生淋巴结转移或远处转移的肉瘤中hTERTmRNA表达有升高趋势(P>0.05);口腔肉瘤中hTERT蛋白的阳性表达率明显高于良性肿瘤(P<0.05),并随肉瘤分化程度的降低而呈升高趋势,其中低度恶性肉瘤与高度恶性肉瘤比较差异显著(P<0.05)。hTERT蛋白表达与肉瘤复发密切相关(P<0.05),发生淋巴结转移或远处转移的肉瘤中hTERT蛋白表达明显升高(P>0.05)。结论:hTERT基因在口腔颌面部肉瘤发生与发展过程中可能起重要作用。
Objective : To investigate the biological significance of the expression of hTERTmRNA and its protein in oromaxillofaeial sarcoma. Methods: In specimens of 37 sarcomas and 14 benign mesenehymal tumor tissue from oral and maxillofaeial region, the expression of hTERTmRNA was determined using in situ hybridization, and hTERT protein was determined by Immunohistochemistry. Results: Compared with benign mesenehymal tumors, the positive rate of hTERTmRNA expression was signifieantly high in oromaxillofaeial sarcomas (62.2%/14.3% , P 〈 0.05 ). The positive rates of hTERTmRNA in sarcomas of low-grade malignaney, moderate grade and high grade were 41.7%, 50.0% and 86.7%, respeetively. There was a significant differenee between sarcomas of low-grade malignancy and high grade ( P 〈 0.05). The expression level of hTRET protein was significantly inereased in sarcomas compared to benign tumors ( P 〈 0.05), and tended to inerease with malignant degree. A significant difference was found between sarcomas of low-grade malignancy and high grade ( P 〈 0.05). hTERT expression and hTERT protein were closely associated with presence of local recurrence ( P 〈 0.05). In sarcomas with distant metastasis and/or lymph node metastasis, the positive expression of hTERTmRNA and hTERT protein tended to increase compared to the other sarcomas ( P 〉 0.05). Conclusion: hTERT gene may play an important role in carcinogenesis and development of oromaxillofacial sarcoma.
