丹参药物血清对HUVEC黏附分子表达及胞内Ca^(2+)的影响 被引量:4

摘要 目的探讨丹参药物血清抗血管内皮细胞凋亡的分子机制。方法取人脐静脉内皮细胞(HUVEC),用脂多糖造成凋亡细胞模型,通过流式细胞仪、激光共聚焦显微镜观察丹参药物血清对ICAM-1表达及胞内Ca2+的影响。结果丹参药物血清可明显降低LPS所致的ICAM-1表达上升,并缓解其所致的钙超载。结论丹参抗HUVEC凋亡并有效防治血栓性疾病的机理与其调控黏附分子ICAM-1表达及缓解细胞钙超载有关。 OBJECTIVE To investigate into the mechanism of the medicated serum of Radix Salviae Miltiorrhizae in preventing endothelial cell apoptosis of in the vessel. METHOD Human umbilicus vein endothelial ceil and lipopolysaccharide were used to establish apoptosis ceil models, and flow cytometer and confocal microscope were used to observe the effect of the medicated serum on ICAM-1 expression and intraceilular Ca^2+. RESULT The medicated serum could markedly check the increase of ICAM-1 caused by LPS and relieve calcium overload. CONCLUSION The mechanism of the effect of Radix Salviae Miltion-hizae in preventing apoptosis and preventing and treating thrombotic diseases is related to its effect in regulating ICAM-1 expression and relieving ceil calcium overload.
