Objective To observe the effects of different doses of simvastatin on plasma vasoaction substances in hypertensive patients. Methods Ninety patients with essential hypertension were randomly separated into three groups. Thirty patients in group A were given routine therapy(ACEI and CCB) ; Thirty patients in group B were given routine therapy and 20 mg simvastatin once daily for 8 weeks ; Thirty patients in group C were given routine therapy and 40 mg simvastatin once daily for 8 weeks. The levels of endothelin, angiotension Ⅱ , nitric oxide, calcitonin gen-related peptide were measured. Results After 8 weeks treatment, nosignificant difference in endothelin, angiotension Ⅱ , nitric oxide and calcitonin gen-related peptide between 20 mg/d simvastatin group and control group were found. However, 40 mg/d simvastatin significantly decreased endothelin and angiotension Ⅱ were markedly increased in nitric oxide and calcitonin gen-related peptide. There was no relation between the decreased magnitude of endothelin, angiotension Ⅱ and that of LDL-C. The decreased in LDL-C was negatively related to nitric oxide and calcitonin gen-related peptide. Conclusion High dosage simvastatin improves the levels of plasma vasoactive substances along with decrease in blood pressure.
