摘要
以杜氏盐藻Dunaliella salina总RNA为模板,根据其它物种MAPK基因的保守序列设计一对简并引物,通过RT—PCR技术扩增获得1条297bp的cDNA片段。再以此片段为模板设计引物,通过RACE技术获得杜氏盐藻MAPK基因的全长序列。杜氏盐藻MAPK基因由664个氨基酸组成,大小为1994bp。在Genebank中进行Blastp检索,发现其与许多物种的MAPK基因具有较高的相似性(55.6%~69.7%)。系统进化分析进一步证明,杜氏盐藻MAPK与衣藻Chlamydomonas reinhardtiiMAPK亲缘关系最近。
The RNA of Dunaliella salina was treated as cyclostyle. The degenerate primers were designed based on the conservative sequence of MAPK from other species. A eDNA fragment of 297 bp from D. salina was obtained by RT--PCR technology. Based on this sequence, a pair of primers was designed. According to RACE technology, the complete MAPK gene was constructed in Dunaliella salina. The whole sequence of MAPK was 1994 bp in size, which encoded 664 amino acids. Blastp search in genebank showed that MAPK of D. salina shared high similarity with that in other species up to 55.6%~69.7%, Phylogenic analysis displayed that MAPK of species in D. salina had a close relationship with those of Chlamydomonas reinhardtii.
