重组腺病毒rAdCDglyES的ES基因生物活性鉴定

摘要 目的:对构建的含融合自杀基因CDglyES的重组腺病毒中的ES基因是否具有独立的生物活性功能进行鉴定,为rAdCDglyES用于治疗恶性肿瘤奠定理论依据。方法:用重组腺病毒rAdCDglyES培养上清,在体外进行内皮细胞ECV-304生长抑制实验,同时做鸡胚绒毛尿囊膜血管生成实验,观察血管生成抑制。结果:重组腺病毒rAdCDg-lyES培养上清对内皮细胞ECV-304的生成有抑制作用(抑制率78.7%),而在鸡胚绒毛尿囊膜血管生成实验中加入培养物上清的实验组与对照组相比鸡胚尿囊膜血管密度稀疏,平均血管数差异明显(P<0.01)。结论:构建的重组腺病毒rAdCDglyES表达产物具有独立的ES功能即抑制内皮细胞增殖,抑制新生血管形成的生物功能。 Objective: To identify whether the ES gene of the recombinant adenovirus containing fusion gene CDglyES constructed by our research group presents independent biological actions and set up the theoretical basis for treating carcinoma with rAdCDglyES. Methods: The growth inhibition test was made in vitro with epithelial cell ECV-304 treated by the supernatant fluid of the recombinant adenovirus rAdCDglyES. Meanwhile, the vessel forming test of chorioallantoic membrane (CAM) of enbryonated eggs was made to observe the growth inhibition of vessels. Results: The supernatant fluid of recombinant adenovirus rAdCDglyES inhibited the growth of epithelial cell ECV-304 (inhibition rate: 78.7%). The average number of vessels was different obviously by comparing the vessel density of CAM of the test group treated by viral supernatant and that of the control group (P〈0.01).Conclusion: The expression product of the recombinant adenovirus rAdCDglyES presents the independent function of ES, that is, the biological function of inhibition of the reproduction of epithelial cells(ECV-304) and the formation of new vessels of CAM.
