摘要
以软枣猕猴桃、黑加仑、越橘、梨、李、杏、小苹果和山定子等果树冷冻幼叶为试材,采用修改的CTAB法提取基因组DNA。利用CTAB提取液提取幼叶组织3次,以第2次提取的DNA产率和质量最好;第3次的质量最好,但产率下降。用第2次和第3次提取的DNA直接用于RAPD分析,重复2次,带型稳定。
Extraction of genome DNA of seven kinds of fruit trees in the north-cast of China via amended CTAB method from young frozen leaves as materials, the splitting leaf tissue was extracted for three times by CrAB buffer. And the result showed that the quality of DNA of the third time was best among the three times while the yield and quality of DNA of the second time were better than other two and the DNA of the second and third time were used for analyses of RAPD PCR directly which bands were stable for two repeats.
