摘要 本文使用价格低廉、研究极少的硅藻土和壳聚糖分别对藻蓝色蛋白粗提液进行了初步纯化,得到了良好的结果。SDS-PAGE电泳测得产品α亚基和β亚基的分子量分别约为16000、19000 u,符合文献报道。藻胆蛋白差示扫描热分析(DSC)结果显示变性温度峰值为56.96,70.32℃,热效应△Hd分别为-1.16和-0.5621 J/g,表明藻胆蛋白亚基间的键能较低,稳定性差。此外,还研究了Cu(Ⅱ),Cr(Ⅵ)对藻蓝蛋白的荧光淬灭作用,对找到一种快捷准确测量某些重金离子浓度的方法具有启发意义。 In this paper, cheap and few researched-diatomitc and chitosan were used for the purification of phycocyanin rough product,through which the positive results were achieved. By the SDS-PAGE, the molecular weights of the two subunits of phycocyanin were assayed to be about 16000 u and 18000 u,respectively, which were dose to the ones report. The result of DSC showed two peak values of denatured temperature resting on 56.96 ℃ and 70.32 ℃ that correspond to the changes in enthalpy (ΔHd) of-1.16 J/g, and-0.5621 J/g, respectively,indicating low bonding-energy between the subunits and bad stability of phyeobiliprotein. The fluorescence quenching of phycocyanin by Cu(Ⅱ) and Ca-(Ⅵ) ions were also studied, which enlightened a new shortcut and accurate way to measure the concentration of some metal ions.
