摘要 目的:探讨脑卒中后抑郁(PSD)大鼠模型建立的可行性、有效性和简便性。方法:采用线栓法制备局灶性脑缺血大鼠模型,并结合孤养、束缚应激制成PSD大鼠模型,观察大鼠糖水消耗试验,自发性行为改变,下丘脑单胺类神经递质的变化。结果:模型组大鼠蔗糖水饮用量与正常组相比明显减少(P<0.01);旷野试验中模型组大鼠水平及垂直得分均小于正常组(P<0.01);模型组大鼠下丘脑NE、5-HT、DA含量均明显小于正常组(P<0.01或0.05)。结论:在局灶性脑缺血大鼠模型基础上结合孤养、束缚应激制成的PSD大鼠模型,可为卒中后抑郁的研究提供较为理想的动物模型。 Objective:To study the feasibility, reliability and utility of the model of poststroke depression(PSD). Methods:A PSD rat model was set up on the base of focal cerebral ischemia by means of middle cerebral artery occlusion, followed by isolated feed and restraint stress. The consumption of sucrose water and the change of animal behavior were observed and the high performance liquid phase- electrochemical was used to detect the biogenic monoamine neurotransmitter content in the hypothalamus area. Results:The consumption of sucrose water of the model rat decreased significantly in comparison to that of the controls (P〈0.01), and the spontaneous behavior of the model rats decreased significantly in comparison to that of the controls (P〈0.01 or 0.05), and the level of NE, 5-HT, DA in the hypothalamus area decreased (P〈0.01 or 0.05). Conclusion:The restrainted rat model set up on the basis of cortical infarction of middle cerebral artery occlusion is an ideal animal model of poststroke depression for further researches.
