摘要
目的了解在体外培养的牛眼小梁细胞(trabecular meshwork cells,TMCs)中缓激肽(bradykinin,Bk)对前列腺素E2(prostaglandin E2,PGE2)信号的影响,探讨两种信号系统在TMCs中相互作用的方式。方法Bk、PGE2单用及联用刺激TMCs,用放射免疫的方法检测细胞内环磷酸腺苷(cAMP)的生成量。结果Bk能够增强PGE2对TMCs的刺激作用;PGE2和Bk联合使用比单用PGE2能更大程度地升高TMCs内cAMP的浓度(P<0.05)。结论Bk可能是通过G蛋白藕联受体(G protein-coupled receptor,GPCR)显著增强PGE2的信号,导致细胞内cAMP的生成显著增多。
Objective To understand the influence of bradykinin(Bk) on the signaling of prostaglandin E2 (PGE2) in cultured bovine trabecular meshwork cells (TMCs), and to discuss the mode of interaction between the two signal pathways in TMCs. Methods TMCs were incubated in 1 000 nmol/L PGE2 or 1 000 nmol/L PGE2 combined with 100 nmol/L Bk for 60 s. The production of intracellular cAMP was assayed by radioimmunoassay. Results Bk could magnify the stimulation of PGE2 on cultured TMCs. PGE2 combined with Bk could cause greater elevation of intracellular cAMP than PGE2 alone(P〈0.01). Conclusion Bk can enhance the signaling of PGE2 through the G protein-coupled receptor, which results in significant formation of cAMP in TMCs.
