摘要
目的 明确空心莲子草类盐抗汉坦病毒(HV)的作用机制。方法 采用噻唑兰(MTT)比色法检测细胞活性、免疫荧光法(IFA)和逆转录聚合酶链反应(RT-PCR)检测病毒滴度和病毒核酸,以确定空心莲子草类盐细胞内抗HV作用。结果 空心莲子草类盐对HV无直接灭活作用,也不能阻止HV的吸附,这表现在各药物浓度组病毒滴度均在5.7以上;空心莲子草类盐有抑制HV生物合成作用,在质量浓度为40-320mg·L^-1内,随着药物质量浓度的增加,病毒滴度、细胞培养液中病毒核酸的阳性稀释度逐渐降低,抗病毒指数逐渐增加,并表现出量效关系,药物治疗组的病毒滴度明显低于病毒对照组(P〈0.05)。结论 空心莲子草类盐在体外通过抑制病毒生物合成而发挥抗HV作用。
OBJECTIVE To investigate anti-hantavirus (HV) activity of compounds obtained from griseb extracts in vitro. METHODS The arlti-HV activities were investigated. Cell activity was determined by MTT colorimetric assay, Viral liters were measured by immunofluorscence assay (IFA) and HV-RNA was detected by reverse transcription polymerase chain reaction (RT-PCR).RESULTS No antiviral abilities of directly-killing and adsorption-bilking were obtained while viral titers were over 5,7 at all concentrations, Anti-biological synthesis was observed at the doses ranging from 40 to 320 mg·L^-1, both viral titers and titers of HV-RNA in culture media decreased correspondingly, whereas the antiviral index increased. The viral titers in the tratment group were significantly lower than that of HV control ( P 〈 0.05). CONCLUSION Griseb may inhibit HV biological synthesis instead of directly inactivating these viruses or blocking their adsorption to susceptible cells in vitro.
