摘要
目的研究重组人白细胞介素1受体拮抗剂(RhIL-1Ra)对大鼠佐剂性关节炎(AA)的影响,探讨部分机制。方法将SD大鼠随机分为6组:正常组、AA组、AA+RhIL-1Ra(2·5、10、40mg/kg,tid,sc)和AA+anakinra组。弗氏完全佐剂诱导AA大鼠模型,检测体重、足爪肿胀度、多发性关节炎指数、T、B细胞增殖度、腹腔巨噬细胞产生的IL-1、TNF-α和PGE2水平及关节滑膜的病理学改变。结果RhIL-1Ra(2·5、10、40mg/kg,tid×7d)改善了AA大鼠踝关节的炎性肿胀和病理学改变,RhIL-1Ra(40mg/kg,tid×7d)显著提高了ConA诱导的脾细胞增殖,RhIL-1Ra(2·5、10、40mg/kg,tid×7d)抑制了LPS诱导的脾细胞增殖,RhIL-1Ra使AA大鼠腹腔巨噬细胞产生IL-1、TNF-α和PGE2减少。结论RhIL-1Ra对AA大鼠有治疗作用,影响腹腔巨噬细胞功能和T、B淋巴细胞可能是其发挥作用的部分机制。
Objective To study the therapeutic effects of recombinant human interleukin 1 receptor antagonist (RhIL-1Ra)on rat adjuvant arthritis (AA) and discuss a part of the mechanism. Methods SD rats were divided randomly into six groups, including normal, AA, AA + RhIL-1 Ra ( 2. 5,10,40 mg/kg, tid) and AA + anakinra (40 mg/kg, tid) groups. Complete Freund's adjuvant was used to induce AA in rats. The effect of RhIL-1Ra on AA rats was measured by paw-swelling, polyarthritis index, splencytes proliferation, activity of interleukin-1 (IL-1), tutour necrosis factor-alpha (TNF-α) and prostaglandin E2 (PGE2 ) production in peritoneal macrophages and histopathological assessment of synovium. Results RhIL-1 Ra (2. 5,10,40 mg/kg, tid × 7 d) subcutaneous injection (sc) inhibited inflammatory swelling, pathologic change in ankle of AA rats. RhIL-1Ra(40 mg/kg, rid, sc ×7d) significantly increased ConA-induced splenocytes proliferation and RhIL-1 Ra (2. 5,10,40 mg/kg, tid, sc × 7 d) supressed LPS-induced splenocytes proliferation. RhIL-1Ra reduced the enhanced production of PGE2, IL-1 and TNF- α from peritoneal macrophages in AA rats. Conclusion RhIL-1 Ra has the therapeutic effect on AA rats, which might be related with its effect on secretory function of activated PM.
