摘要 目的:了解房水中细胞因子对体外培养牛眼小梁细胞合成富含半胱氨酸的酸性分泌蛋白(secretedprotein,acidicandrichincysteine,SPARC)的影响。方法:体外培养牛眼小梁细胞并传3代,应用免疫组化染色法验证SPARC存在于小梁细胞内,并将传3代的小梁细胞无血清培养24h后分别加入房水中的细胞因子bFGF,TGF-β,EGF及IGF-1,72h后提取细胞上清液,并用酶联免疫吸附法(ELISA)测定SPARC的浓度。结果:细胞因子bFGF,TGF-β不影响小梁细胞对SPARC的合成,EGF和IGF-1分别减少和增加小梁细胞对SPARC的合成。结论:细胞因子可通过影响SPARC的生成而调节眼内压。 AIM: To evaluate the effect of cytokine in aqueous humor on secreted protein, acidic and rich in cysteine (SPARC) synthesis by bovine trabecular cells (BTCs). METHODS: BCTs were primarily and subcultured, then anti-SPARC antibody was used for immunohistochemical stains of the cultured cell. The third passage cells were cultured for 24 hours on serum-free medium, and then cultured for 72 hours on serum-free medium with bovine basic fibroblast growth factor (bFGF), transforming growth factor-β (TGF-β), epidermal growth factor (EGF), or insulin-like growth factor-1 (IGF-1). SPARC concentration in the medium was measured by the ELISA method. RESULTS: The addition of bFGF or TGF-β did not affect SPARC synthesis by the BCTs. However, the synthesis of SPARC by BCTs decreased by addition of EGF and increased by addition of IGF-1, respectively.OONCLUSION: Cytokine in aqueous humor may regulate intraocular pressure by affecting the synthesis of SPARC.
