摘要
目的建立一种能有效地评价小鼠恐惧记忆能力的实验动物模型。方法选用昆明种(KM)和C57BL/6小鼠,使用声音结合足底电击训练,测试同一品种小鼠用不同电击强度,和不同品种小鼠用相同条件训练时,恐惧性记忆建立的差异。恐惧性记忆形成的表现为训练后24h测试时出现木僵反应(stuporresponse)且持续时间长。结果声音分别结合0.4mA、0.6mA、0.9mA的足底电击,24h后在KM小鼠引出的木僵反应时间分别为(60.00±57.77)s,(110.00±129.61)s,(101.99±20.38)s,组间没有显著性差异(P>0.05)。强度0.6mA组的合格率最高,达30%。在声音结合0.4mA电击条件下,C57BL/6小鼠的木僵反应时间为(275.62±36.99)s,显著长于KM小鼠的时间(P<0.01);前者的合格率为100%,远高于KM小鼠的7.69%。结论C57BL/6小鼠比KM小鼠更适用于建立恐惧记忆模型;强度0.4~0.6mA的足底电击能获得重复性好的训练效果,而仅以增加电击强度的方式不能提高合格率。
Objective To establish an optimal fear conditioning protocol for studying the inter-strain difference in mice. Methods Kunming mice (KM) and C57BL/6 mice were examined. The conditioning was to associate a footplate shock with a tone signal. The duration of tone-induced stupor response 24h after the training represents the retention of fear memory. Results In both strains the optimal current intensity inducing a reproducible response was 0.4 - 0.6mA. In KM trained using 0.4, 0.6, and 0.9mA aversive stimuli, the durations of stupor response were 60.00 ± 57.77 s, 110.00 ± 129.61 s and 101.99 ± 20.38 s respectively( P〉 0.05 ) ; the highest qualification percentage (30%) was seen in 0.6mA group. Using 0.4mA aversive stimulus, the duration was 275.62 ±36.99s in C57BL/6, which was significantly than that in KM ( P〈0.01 ). All C57BL/6 mice qualified, while only 7.69% in KM( P〈 0.01 ). Conclusion C57BL/6 strain is more suitable for fear conditioning studies than KM strain and current of 0.4 -0.6 mA is optimal intensity.
