吸烟对外周血单个核细胞表达烟碱乙酰胆碱受体的影响

摘要 目的研究健康男性外周血单个核细胞(PBMCs)烟碱型乙酰胆碱受体(nAchR)亚型的表达情况及吸烟对其表达量的影响。方法抽取92例健康男性吸烟者和36例不吸烟者的外周静脉血,通过半定量RT-PCR对其nAchR11种亚基(α2~α7、α9、α10、β2~β4)mRNA在PBMCs的中表达情况进行分析。结果11种nAchR亚基只有α2、α5、α6、α7四种亚基mRNA在吸烟者和不吸烟者PBMCs中有表达,吸烟者α5、α7亚基mRNA的表达量比不吸烟者低(均P〈0.01),而两者α2亚基mRNA的表达无显著性差异(P〉0.05)。结论烟碱与外周血免疫细胞上nAchR的相互作用可能是吸烟导致机体免疫系统功能改变的因素之一。 Objective To investigate the expression of nicotinic acetylcholine receptor (nAchR) subunits, and the effects of smoking on its expression in peripheral blood mononuclear cells (PBMCs). Methods Blood was collected from 92 healthy male smokers and 36 healthy male non-smokers. Peripheral blood monocytic cells were obtained, and reverse transcription-polymerase chain reaction (RT-PCR) was employed to study the gene expression of 11 nAchR subunits (α2-α7, α9, α10, β2-β4). Results The results showed that the PBMCs of smokers and non-smokers appeared to express α2, α5, α6 and α7 only out of 11 nAchR subunits. However, the expression of α5 and α7 subunits was lower in smokers' PBMCs than in non-smokers' (P〈0.01), and showed a dose-dependent relationship with the daily cigarette consumption (P〈0. 01). Conclusion It is suggested that the nAchR may be expressed in human PBMCs. Smoking appears to decrease the expression of α5 and α7 subunits. The authors presumed that smoking might bring about a certain effect on the immune system through the interaction of nicotine with nAchR on PBMCs.
