摘要 目的:建立检测患者全血中环孢素A(CsA)浓度的高效液相色谱法。方法:血样经乙醚-石油醚(70:30)萃取后,以C18硅胶键合相为固定相,乙腈-甲醇(80:20)为流动相,70℃柱温下进行色谱分析,紫外检测波长为210nm。结果:CsA全血浓度在50-1600ng/mL范围内呈良好的线性关系,r=0.9998,方法的平均回收率为97.6%,日内和日间精密度RSD均小于6%(n=5),最低检测限为10ng/mL。结论:本法简便、快速,易于开展,用于肾移植术后患者CsA的血药浓度监测,效果良好。 Objective:To establish a practical RP-HPLC method to determine the concentration of eyclosporin A in patient's whole Hood. Methods: CsA was extracted from whole blood with ether and separated, using a C18 column and a mobile phase of acetonitrile-methanol (80:20). The column temperature was maintained at 70 ℃ and CsA was detected at 210 nm. Results: A good linear relationship was obtained between the concentration of CsA from 50 ng/mL to 1 600 ng/mL, r = 0.9998. The average recovery rate of CsA was 97.6%, and the RSD within a day and between days were less than 6% (n = 5). The detection limit of CsA was 10 ng/mL. Conclusions: The established method was accurate, simple, sensitive and suitable for the therapeutic monitoring of CsA.
