摘要
目的当一盒电化学发光试剂用完规定的检测量后,剩余1/3的死腔量试剂一般会被丢弃,若能将其再应用,将会节省1/4的试剂费用。本文探索死腔量试剂再应用的方法并验证其检测能力。方法将三个相同批号的原装CA199试剂盒中的死腔量试剂分别集中至其中一个试剂盒,成为一盒混合试剂。把混合试剂瓶上E lecsys 2010/1010专用条形码数字以手工法输入Modu lar E170,可被Modu lar E170识别,作为一盒新的同批号试剂使用。每盒原装试剂及混合试剂使用时,常规做正常水平和高水平室内质控,用独立样本t检验分析原装试剂与混合试剂的质控数据。结果原装试剂与混合试剂的正常水平质控数据方差齐性,t=0.247,P=0.806;原装试剂与混合试剂的高水平质控数据方差齐性,t=1.274,P=0.206,原装试剂与混合试剂的检测能力无差异。结论电化学发光试剂死腔量的再应用,方法简单可行,检测能力与原装试剂相同,可节省1/4的试剂费用,值得推荐。
Objective When the prescriptive test numbers of a packet reagents for electrochemiluminescence are used up, about 1/3 dead volume reagent remain in the packet which is used to be discarded. If we could think of Ia way to reuse these dead volume reagents, 1/4 reagents expenses may be reduced. The aim of this study is to research for the way of reapplication and to assess the efficacy of dead volume reagents. Methods Collect the dead volume reagents of three same lot number packets of CA 199 together to from a mixed reagents packet. There are barcode figure for Elecsys 2010/1010 on mixed reagent packet. We manually registrate the figure into Modular E170, then the mixed reagents packet can be accepted as a prototype reagents packet with same lot mumber. When per mixed reagents packet is applied, normal level and high level quality control are routinely performed, as is done on prototype reagents packet. We statistically analyze the data by independent sample test. Results With normal level quality control of prototype reagent and mixed reagent, the t value is 0. 247, and the P value is 0. 806. With high level quality control of prototyep reagent and mixed reagent, the t value is 1. 274 and the P value is 0. 206. There is no significant difference in the ablility to detect CA 199 between mixed reagents and prototype reagents. Conclusion The ability of mixed reagent packet to detect CA 199 is same to that of prototype reagent packet. The reapplication of dead volume reagents for electrochemiluminescence is feasible and convenient, and may reduce 1/4 reagents expenses.
