镓盐对人胃癌MGc_(80-3)细胞DNA合成的影响及其抑瘤机制

摘要 采用细胞培养, ̄3H-TdR同位素示踪及MTT显色技术观察了氯化镓(GalliumChloride,GC)对人胃低分化粘液腺癌MGc_(80-3)细胞株的生长抑制作用及DNA合成的影响,并以铁为实验因子探讨镓抑瘤作用机制。结果表明:稼能使MGc_(80-3)人胃癌细胞 ̄3HTdR掺入显著减少,抑制癌细胞DNA合成。镓不仅使癌细胞生长抑制,还可使癌细胞死亡。这种抑制作用可被Fe ̄(+3)的加入而减弱。提示:GC抑制癌细胞生长的机制可能与癌细胞铁利用障碍致DNA合成低下有关。 This study, using gastric carcinoma cell culture,  ̄3H-TdR isotope incorporation and MTT assay technique, observed the growth inhibition effect of gallium chloride(GC)on poorly differentiated mucoid adenocarcinoma of human stomach MG_(c80-3) cell line and the effect of GC on DNA synthesis. Using FeCl3 as an objection, we investigated the mechanism of the growth inhibition effect of gallium on MG_(c80-3) cell.The result suggested that  ̄3H-TdR isotope uptake was markedly decreased by GC,and GC could inhibit DNA synthesis.GA_(+3) can not only make cancer cell grow inhibitively,but also make the cell die.This inhibitive effect can be decreasedby Fe_(+3).The result indicates that the mechanism of GC inhibiting cancer cell may be related with the block of uitilizing Fe in cell, which results in decrease of DNA synthesis.
