果糖诱导高血压伴胰岛素抵抗大鼠血清脂联素和肿瘤坏死因子α水平的变化

摘要 目的:建立高血压伴胰岛素抵抗动物模型并观察血清脂联素、肿瘤坏死因子α的变化。方法:实验于2005-07/09在海军总医院中心实验室完成。选择6周龄雄性Wistar大鼠20只,随机数字表法分为两组,对照组(n=10)以普通标准饲料+普通自来水喂养i果糖组(n=10)以普通标准饲料+10%果糖水喂养,连续喂养14周,于喂养后第8和14周末,采用尾部测量法测定大鼠血压,胰岛素抵抗指标采用胰岛素敏感性指数=-ln[空腹血糖×空腹胰岛素],采用放射免疫法测定血清脂联素和肿瘤坏死因子α的浓度。结果:纳入动物20只,均进入结果分析。①第8周末果糖组肿瘤坏死因子α水平与对照组相比明显升高[分别为(2.21±0.34),(1.84±0.25)μg/L,P<0.05],胰岛素敏感性指数、脂联素明显下降[分别为-4.64±0.12,-4.23±0.27;(4.76±0.67),(5.83±0.99)mg/L,P<0.01]。②第14周末两组间胰岛素敏感性指数、脂联素和肿瘤坏死因子α的差异更加明显,第14周末果糖组胰岛素敏感性指数、脂联素与第8周末相比明显降低[-5.05±0.32,-4.64±0.12;(3.60±0.50),(4.76±0.67)mg/L,P<0.01],肿瘤坏死因子α明显升高[分别为(2.53±0.39),(2.21±0.34)μg/L,P<0.01]。③相关性分析表明,脂联素与空腹胰岛素、胰岛素敏感性指数和肿瘤坏死因子α水平的相关性显著(r=-0.564,-0.641,-0.538,P<0.05)。结论:10%果糖水喂养的Wistar大鼠,具有典型的高血压伴胰岛素抵抗的特点;胰岛素敏感性降低的同时伴有血清脂联素下降和肿瘤坏死因子α的升高。 ATM: To establish rat models of hypertension and insulin resistance, and study the changes of serum adiponectin and tumor necrosis factor (TNF) α. METHODS: The experiment was conducted in the Central Laboratory of Naval General Hospital from July to September 2005. Twenty Wistar male rats of six-week-old were selected and divided into two groups: control group (n=10) and fructose group (n=10). Rats in the control group were fed with standard diet and normal water and rats in the fructose group were fed with standard diet and 10% of fructose water. Rats in both groups were fed for 14 continuous weeks, and the blood pressure was detected at 8 and 14 weeks after feeding with tail-cuff method. The insulin sensitivity index (ISI) was taken as the insulin resistance index [ISl=-In (fasting blood glucose xfasting insulin)]. The level of serum adiponectin and TNFα were determined by radioimmunoassay RESULTS: A total of 20 enrolled rats were involved in the analysis opf resuhs.(1)The level of TNFα at the 8^th week was obviously increased in the fructose group than the control group [respectively (2.21±0.34), (1.84±0.25)μg/L,P 〈 0.05], while the ISI and serum adiponectin in fructose group were significant decreased [-4.64±0.12,-4.23±0.27;(4.76±0.67), (5.83±0.99) mg/L,P 〈 0.01]. (2)The differences in ISI, adiponeetin and TNF α between two groups at the 14^th week were more significant. The ISI and level of adiponeetin in the fructose group was significantly decreased at the 14^th week than the 8^th week [-5.05±32,-4.64±0.12; (3.60±0.50), (4.76±0.67) mg/L,P 〈 0.01], while the level of TNFα was significantly increased [respectively were (2.53±0.39), (2.21±0.34)μg/L,P 〈 0.01]. (3)Correlation analysis showed that adiponeetin was significantly related to fasting insulin, ISI and level of TNF α(r=-0.564,-0.641 ,-0.538,P 〈 0.05 ). CONCLUSION: Rats fed with 10% of fructose water are characteristic of typical hypertensio
