摘要 以中国春缺体-四体系为材料,提取各材料的基因组DNA,分别经限制性内切酶EcoRI和KpnI完全酶切、电泳、转膜后,用α-32P_dCTP标记的与小麦耐盐相关的糖原合成酶激酶基因TaGSK1为探针进行杂交,结合生物信息学的方法,将该基因定位于普通小麦第一同源群染色体的短臂上,为进一步研究小麦耐盐性的遗传及机理奠定了基础。 In this paper, Chinese Spring null-tetrasomic lines was studied as material. Their genomic DNA were extrated and were digested completely by EcoRI and KpnI. After electrophoresis and transmembrane, digested genomic DNA hybrided with the probe of the gene of glycogen synthase kinase 1 in Triticum aestivum ( TaGSKI ), involved in salt-tolerance and marked with a-32p-dCTP. Associated with Bio-information, TaGSKI was located in the short ann of 1st chromosom group, ie. 1AS, 1BS and 1 DS. This paper established the base of heredity and mechanism in wheat salt-tolerance.
