摘要
目的探讨肝癌(H22)超声固化瘤苗在高压氧环境下诱发小鼠的抗肿瘤效应及对免疫功能的影响。方法将实验小鼠随机分为对照组、常压瘤苗组和高压氧瘤苗组,采用肝癌(H22)超声固化瘤苗对小鼠进行免疫实验,通过癌细胞攻击实验和胸腺细胞自发性3H-TdR掺入量测定,观察其在小鼠体内诱发的抗瘤和胸腺免疫效应。结果A组的抑瘤率为0、B组和C组的抑瘤率分别为83.33%和91.67%,死亡率分别为16.67%和8.33%,生命延长率分别为75%和87%,6周生存天数(39.14±5.6)d和(51.28±3.7)d,肿瘤发生率分别为41.66%和25%,其两组与A组比较差异有统计学意义(P<0.05),而B组和C组比较差异也有统计学意义(P<0.05)。免疫B组和C组的腺细胞平均数显著上升,而胸腺细胞3H-TdR掺入量明显下降。结论免疫小鼠可获得对随后接种同源细胞攻击,有免疫保护作用,并产生免疫记忆;高压氧曝露的免疫小鼠的抗肿瘤效应和免疫力明显强于另外两组,超声固化瘤苗与高压氧结合能显著地提高激活宿主免疫系统,抑制肿瘤生长和延长生命,有望成为一种新的、安全的、有效和无毒副作用的综合治疗新模式。
Objective To investigate the anti-cancer results and immunizing effect of ultrasound solidification tumor vaccine of mouse in hyperbaric oxygen circumstance. Methods The hundreds of mouse randomization into control group, ordinary pressure tumour vaccine group, hyperbaric oxygenation ultrasound solidification tumor vaccine group. The immunizing test was taken on ultrasound solidification tumor vaccine circumstance. Results The total remission rates of tumor in A group, B group and C group were 0%, 83.33% and 91.67% respectively. The mortality rates were 16.67% and 8.33% in B group and C group respectively. The life span prolongation rates were 75%, 87% respectively. The mean survival time were ( 39.14±5.6)d, (51.28±3. 7)d respectively. The tumorigenesis rates were 41.66 % and 25 % respectively. The B, C group vs A group the difference was remarkable in statistics (P〈0.05). The number of immunizing glandular cell increased remarkable in B group and C group. The :^3H-TdR incorporate rate of thymus cell decreased obviously. Conclusion The immunized mouse may have the ability of attack the late inoculated isogenic cell. In other words, It can get immunoprotection and produce immunological memory. The immunity and anti-cancer ability of immunized mouse on hyperbaric oxygen condition better than those of the other groups. The combination of hyperbaric oxygen therapy with ultrasound solidification tumor vaccine get better therapeutic effect and lower side effects than the control group. It may activate the host immunologic system. It is to be worth to generalization.
