摘要
目的采用乙酸法制备大鼠溃疡性结肠炎(UC)模型,观察培菲康(bifico)对UC的治疗效果并探讨其作用机制。方法用8%乙酸灌肠法制备大鼠UC模型,给大鼠应用bifico溶液,观察UC大鼠的症状和组织学变化,并采用原位杂交技术,检测UC大鼠肠黏膜内IL-1β、TNF-α、IL-10、NF-κB的表达水平。结果应用bifico后,UC大鼠的症状、组织损害均较模型组明显减轻,同时IL-1β、TNF-α、NF-κB的表达明显降低,IL-10的表达明显提高。结论bifico可有效治疗UC,机制可能是通过抑制NF-κB的活性,进而降低致炎细胞因子的表达,提高抗炎细胞因子的水平而完成的。
Objective To investigate the effects of probiotics on the expression of cytokines and nuclear factor kB in intestinal mucosa of rats with ulcerative colitis(UC). Methods Rat model with UC was established by enema with acetic acid. Rats of UC were treated with probiotic solution for 7 days. The expression of cytokines in the intestinal mucosa of rats were detected by in situ hybridization technique. Following oral probiotics the exprexsion of cytokines and NF-kB was analyzed. Results Compared to control rats, symptoms and lesions of colonic mucosa were improved in rats with oral probiotics. Futhermore, the expression of TNF-α, IL-1β and NF-kB was significantly decreased(P〈0.05),and the expression of IL-10 was increased. Conclusion Probiotics have therapeutic effects on UC. It may suppress the activity of NF-kB to inhibit the expression of proinflammatory cytokines.
