摘要
目的:探讨神经生长因子(NGF)在疼痛发生机制中的作用。方法:采用大鼠后足掌皮下注射福尔马林溶液的外周炎性痛模型。24只成年SD大鼠随机分为4组(每组6只),包括对照组(A组)、致痛后2h取材组(B组)、致痛后24h取材组(C组)和致痛后48h取材组(D组)。观察致痛1h内疼痛行为变化,应用免疫组织化学方法结合计算机图像分析技术检测致炎足底皮肤、背根神经节(DRG)和脊髓背角神经元内NGF的表达,及DRG和脊髓背角神经元表达P物质(SP)的变化。结果:致痛组大鼠都表现出明显的两期伤害性反应。NGF免疫阳性反应物在所有致痛组患侧足底皮肤和DRG表达都增加,NGF和SP免疫阳性反应物在C组患侧脊髓背角表层的表达增加。结论:NGF不仅是产生炎性痛外周机制的重要介质之一,也可能具有中枢作用机制。
Objective: To determine the effect of nerve growth factor (NGF) in the pain mechanism. Methods: Peripheral inflammatory pain model was produced by an postpedes subcutaneous injection of formalin into rats.Twenty-four adult SD rats were randomly divided into four groups (each group contained six rats) including the control group (group A),two hour after formalin injection (group B ), twenty-four hours after formalin injection (group C) and forty-eight hours after formalin injection (group D) . Pain response was observerd into one hour then NGF expression in the inflammatory skin of sole ,dorsal root ganglion (DRG) and spinal dorsal horn neuron and SP expression in the DRG and spinal dorsal horn neuron were evaluated by immunochemistry technique and computer image analysis methods. Results: There were significant bi-phasic nociceptive response of formalin-induced in rats.The expression of immune positive reaction in NGF all increased in inflammatory sole skin and the expression of NGF and SP immune positive reaction increased in the spinal dorsal horn neuron of the group C. Conclusion: NGF is not only an important transmitter in the peripheral machenism of inflammatory pain, but also contribute to central effect mechanism.
