Establishment of Plant Regeneration of Populus euramericana San Martino

摘要 以水培I-72杨(Populus euramericanaSan Martino)的叶片和叶柄为外植体,探讨了I-72杨植株再生的方法。结果表明:I-72杨愈伤组织诱导和不定芽分化的最佳培养基为MS+BA 0.5 mg/L+NAA 0.1 mg/L,愈伤组织诱导率为97.0%,不定芽分化率为91.1%;在诱导培养基中添加Vc(150 mg/L)能有效地抑制叶片的褐变,褐变降低率达25.0%;不定芽在MS+BA 0.3 mg/L+NAA 0.05 mg/L培养基中继代增殖系数为6.3;添加0.8 mg/L GA3对继代培养中不定芽的伸长和增殖有显著效果;在生根培养基1/2 MS+NAA 0.05 mg/L+IBA0.2 mg/L上,生根率达93.5%。 The regeneration system of Populus euramericana San Martino was established. Leaves and petioles sprouted from the cutting shoots cultured in water were selected as explants. The results showed that on the MS medium appended with 0. 5 mg/L BA and 0. 1 mg/L NAA,the ratio of callus inducement was 97.0% and the ratio of bud inducement was 91.1%. The addition of 150 mg/L Vc to the medium could reduce the leaf browning efficiently with the decrease of browning rate of 25. 0%. The proliferation coefficient was 6. 3 after inoculating buds of P. euramericana San Martino into the medium -MS+BA 0.3 mg/L +NAA 0.05 mg/L. Adding 0.8 mg/L GA3 into the medium was apparently favorable to the subculture multiplication of P. euramericana San Martino. The medium for rooting was 1/2 MS+NAA 0.05 mg/L+IBA 0. 2 mg/L and the ratio of rooting was 93. 5%.
