摘要
目的观察脑血灵颗粒对脑出血大鼠脑细胞凋亡及Bcl-2、半胱氨酰天冬氨酸特异性蛋白酶-3(Cas-pase-3)表达的影响。方法以自体血制作大鼠脑出血模型,实验组给予大剂量脑血灵颗粒,华佗再造丸为阳性对照组,模型对照组和假手术组给予生理盐水,通过TUNEL法检测原位凋亡细胞,免疫组化、原位杂交、Western-blot检测Bcl-2、Caspase-3等凋亡相关基因的表达。结果在脑出血造模后7d,TUNEL阳性细胞数明显增多,Bcl-2表达明显下降,Cas-pase-3表达明显上升;脑血灵颗粒剂及华佗再造丸均能使细胞凋亡的数量减少,同时增加Bcl-2的表达,降低Caspase-3的表达;脑血灵颗粒剂组与华佗再造丸组比较,TUNEL阳性细胞数、Bcl-2表达的降低幅度及Caspase-3表达的上升水平,均有统计学意义(P<0.05)。结论脑血灵颗粒具有显著的抗凋亡活性,从而对脑出血后的继发性神经元损伤具有保护作用。
Objective To study the effect of Noaoxielin granule on apoptosis and expressions of Bcl - 2, Caspase - 3 after intracerebral hemorrhage in rat. Methods Male Sprague- Dawley rats received an infusion of 50 μL autologous blood into the right basal ganglia. The rats were randomly divided into sham operated group, model group, Huatuozaizao bolus group and Naoxielin granule group. TUNEL staining were used to detect the expression of apoptosis. The expression of Bcl- 2, Caspase- 3 were observed by immunohistochemistry, in situ hybridization and Western - blot. Results After ICH, Caspase - 3 and TUNEL positive cells increased obviously compared with the sham operat- ed grbup. Bcl- 2 decreased markedly. Both Caspase- 3 and TUNEL positive cells after hemorrhage were significantly reduced by treatment with Naoxielin granule compared with the Hua tuo zai zao bolus group. On the contrary, Bcl- 2 decreased increased markedly. Conclusion Increase Naoxie lin granule might obviously inhibit the activatity of Caspases - 3, increase the activity of Bcl - 2, reduce the number of apoptotic cell after ICH in hemorrhagic brain damage.
