急、慢性白血病患者化疗后胸腺近输出功能状态的研究

摘要 目的研究急、慢性白血病患者化疗后胸腺近输出功能,评价白血病化疗后T细胞免疫状态。方法通过real-time PCR检测56例急、慢白血病患者化疗后外周血T细胞受体重排删除环(TREC)含量,同时检测50例正常健康人TREC水平以确定正常值范围。结果正常人TREC含量为3351.06±3711.12拷贝/105细胞,并且正常人TREC含量与年龄呈负相关。37例急性白血病患者化疗后TREC含量为307.87±433.26拷贝/105细胞;19例慢性粒细胞性白血病患者化疗后TREC含量为314.23±868.63拷贝/105细胞。结论正常人外周血TREC含量与年龄呈相关。急、慢性白血病患者化疗后TREC含量均远远低于正常人水平(P<0.001),但两组患者之间无明显差异(P>0.05)。急、慢性白血病患者化疗后胸腺近输出功能低下,T细胞免疫缺陷。 Objective To analyze the function ofthymic output and to evaluate T cell immune status in acute and chronic leukemia patients after intensive chemotherapy.Methods Real-time quantitative PCR detection of T cell receptor excision DNA circles (TREC) in DNA of peripheral blood mononuclear cells from 56 leukemia patients. The content of TREC in 50 normal individuals also detected and determined the normal range of TREC in health groups. Results The mean value of TREC in normal individuals is 3351.06±3711.12 copies/10^5 cells. There are inverse correlation between TREC and age in health groups. 307.87 ± 433.26 copies/10^5 cells in 37 acute leukemia - patients and 314.23 ±868.6 copies/10^5 cells in 19 chronic leukemia patients, respectively.Conclusions There are inverse correlation between TREC and age in health groups.The contents of TREC in leukemia patients were far. lower than in normal individuals(P〈0.001).There was no difference between two group patients(P〉0.05).The function of thymic output is lower and existence the deficiency of T cell immune in acute and chronic leukemia patients after intensive chemotherapy.
