摘要
目的 研究Notch-1在人脑胶质瘤中的表达,并对其表达与胶质瘤病理级别的关系进行研究,探讨Notch-1对人脑胶质瘤发生、发展所起的作用,从而为胶质瘤的治疗提供一定的理论依据。方法应用半定量RT—PCR方法检测70例人脑胶质瘤标本和12例正常脑组织中的Noreh-1 mRNA的表达。结果Notch-1mRNA在人脑胶质瘤和正常脑组织中均可表达,且在人脑胶质瘤中的表达显著高于正常脑组织(P〈0.01);人脑胶质瘤Ⅰ~Ⅱ级组和Ⅲ~Ⅳ级组中Notch-1 mRNA表达均显著高于正常脑组织(P〈0.01),并且Notch-1 mRNA在人脑胶质瘤Ⅲ~Ⅳ级组中的表达显著高于Ⅰ~Ⅱ级组(P〈0.01)。结论Notch-1 mRNA在人脑胶质瘤和正常脑组织中均表达,且在人脑胶质瘤中呈显著高表达,可能与人脑胶质瘤的发生、发展有关;Notch-1的表达与人脑胶质瘤的病理分级相关。
Objective To study the expression of Notch-1 in human gliomas and the correlation between the expression and the pathological grades of gliomas,to explore the role of Notch-1 in the tumorigenesis and progression of human gliomas, accordingly to provide some proof of theory for treatment of human gliomas. Methods The expression of Notch-1 mRNA was detected in 70 primary human gliomas and 12 normal cerebral tissuses by using RT-PCR semi-quantity method. Results The expression of Notch-1 mRNA have been detected in human gliomas and normal cerebral tissuses and the expression level of Notch-1 mRNA in human gliomas was considerably higher than those in normal cerebral tissuses(P〈0. 01) ;the expression level of Notch-1 mRNA in gliomas of grade Ⅰ-Ⅱ and Ⅲ-Ⅳ was considerably higher than those in normal cerebral tissuses (P〈0. 01 ), and the expression level of Notch-1 mRNA in gliomas of grade Ⅲ-Ⅳ was considerably higher than those in gliomas of grade Ⅰ-Ⅱ. Conclusion The expression of Notch-1 mRNA have been detected in human gliomas and normal cerebral tissuses,and the expression of Notch-1 mRNA in human gliomas was considerably high, Notch-1 may be involved in the tumorigenesis and progression of human gliomas; the expression of Notch-1 may be correlation to the pathological grades of human gliomas.
