摘要
目的观察还原型谷胱甘肽对大鼠肺缺血再灌注(IR)后细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子α(TNF-α)蛋白表达的影响。方法72只雄性清洁级SD大鼠随机分为3组:假手术组(S)、缺血再灌注组(IR)、药物治疗(RG)组。每组分别于夹闭缺血的第45min、缺血再灌注后1、2及4h四个时间点,处死大鼠,取肺组织测定肿瘤坏死因子(TNF-a)、髓过氧化物酶(MPO)含量,免疫组化染色检测肺组织细胞间黏附分子(ICAM-1)蛋白表达。结果TNF-a、MPO含量在IR组缺血再灌注后含量明显增加,较S、RG组显著增高(P<0.01);IR组ICAM-1蛋白表达明显高于S组和RG组。结论还原型谷胱甘肽能减少肺组织TNF-a、I-CAM-1表达,在一定程度上减少肺损伤。
[Objective] To investigate the effect of reduced glutathione intercellular adhesion molecular-1 (Ⅰ- CAM-1), tumor necrosis factor alpha (TNF-α) expression following lung ischemia-reperfusion (IR) in rats. [Methods] Seventy-two SPF SD rats were divided into three groups: sham operation group(S), ischemia-reperfusion group (IR) and reduced glutathione group (RG). Six rats were put to death at one of four time points (45 min ischemia, 1, 2 and 4 hour reperfusion) in each group and taken the left lung. The tumor necrosis factor alpha (TNF-α), myeloperox- idase (MPO) concentration in lung tissue, the intercellular adhesion molecule-1 (ICAM-1) expression in pulmonary endothelium was assessed by histochemical staining. [Results] The TNF-α, MPO concentration in lung tissue: in IR group were increased more obviously than in the S and RG group (P 〈0.01). ICAM-I in IR group expression were increased more obviously than in the S and RG group P 〈0.01) after ischemia- reperfusion. [Conclusion] Reduced glutathione could decrease TNF-α concentration and expression of ICAM-1 in lung tissue after ischemia-reperfusion and could protect lung ischemia-reperfusion injury.
