摘要
目的:建立一种测定金枪鱼鱼片中碳氧肌红蛋白含量的方法。方法:用磷酸缓冲液将肌红蛋白提取出来,加入联二亚硫酸钠将其还原成由脱氧肌红蛋白(Mb)及碳氧肌红蛋白(MbCO)组成的双组分系统,用分光光度计进行比色。根据Mb及MbCO的光学性质,利用比尔定律,求出碳氧肌红蛋白的含量。结果:得出了金枪鱼鱼片中MbCO含量的计算公式。重复测定同一样本,变异系数为2.0%。将实测值与标准值做相关分析,相关系数为0.9998。结论:该法测金枪鱼鱼片中MbCO含量准确、可靠,适用于一般的常规测定。
A analysis method is developed to determine the content of carboxymyoglobin in tuna fillet. After extraction by phosphate buffer, the myoglobin is converted to deoxymyoglobin and carboxymyoglobin by sodium dithionite,and the quantification is done by reading its absorbance against phosphate buffer . According to the different absorbance of Mb and MbCO, the content of carboxymyoglobin is calculated based on Lambert-beer law. Results show that a correlation coefficient of 0.9995 can be obtained between measured level and standard level of carboxymyoglobin, and the coefficient of variation is 1.9%. It's concluded that this method is reliable and precise. It is suitable for routine measurement of carboxymyoqlobin in tuna fillet.
