摘要
目的研究葛根素对心肌细胞H9C2一氧化氮的影响。方法eNOS蛋白用Westernblot方法检测,eNOS基因用RT-PCR方法检测,并测定一氧化氮含量。结果在葛根素2mg/ml浓度下作用24h,可明显提高心肌细胞H9C2eNOS蛋白和基因表达。结论葛根素在心血管方面的保护作用很可能与调节一氧化氮含量及其合酶表达有关。
Objective To investigate the effect of puerarin on the production of nitric oxide in the rat cardiac muscle cell line H9C2. Methods Analysis with western blot and RT-PCR for eNOS protein, eNOS mRNA respectively and measure the NO release. Results It revealed that 2 mg/ml puerarin can elevate the level of endothelial NOS protein and mRNA after 24 h treatment. Conclusion The effect of puerarin on protection of cardiovascular disorders may be due to the regulation of NO production and eNOS.
