摘要 利用聚丙烯酰胺凝胶电泳(SDS-PAGE)对鲤鱼、草鱼、白鲫鱼、鲢鱼、罗非鱼及鲨鱼的肌肉蛋白做鉴别分析.实验采用4种不同抽提液对肌浆蛋白和肌原纤维蛋白进行提取.肌肉蛋白经电泳后以银染色或考马斯亮蓝染色并对蛋白质图谱作对比分析.根据蛋白质条带的数量、迁移率及浓度等特点,得出各鱼种间的相似性及差异性.结果表明,以SDS-PAGE电泳法可实现对水产食品原料品种的初步鉴别. Sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was applied as an effective method in fish species identification. Six fish species, namely, carp, grass carp, crucian carp, silver carp, tilapia and shark were used. A comparison was made among protein extracts from sarcoplasmic, myofi- brillar, SDS-soluble, and urea-soluble fractions, respectively. Species identification of all tested fish could be preliminarily achieved by judging the protein patterns, including the number of bands, mobility and relative concentration on SDS-PAGEs stained by silver or Coomassie brilliant blue R-250.
