摘要
目的:研究脂肪源性内皮细胞向脂肪细胞的分化潜力和过程。方法:获取4只雄性SD大鼠鼠蹊部皮下脂肪,通过机械分割和组织消化法获取脂肪基质细胞,在内皮细胞培养液中培养7d后,经免疫细胞化学及透射电镜鉴定证实95%以上细胞已具内皮细胞表型特征,再将这些细胞进行成脂(DMEM/F-12培养基内加入0.5mmol/L1-甲基-3-异丁基-黄嘌呤,1μmol/L地塞米松,10μmol/L胰岛素,200μmol/L吲哚美星)定向诱导7d,形态学观察和油红O特殊染色鉴定诱导后的细胞。结果:SD大鼠脂肪源性内皮细胞呈典型的铺路石样单层融合贴壁生长;Ⅷ因子、W-P小体阳性;在此基础上经成脂诱导,发现约85%内皮细胞转化为脂肪细胞,油红O染色可见细胞质中有大量脂滴形成。结论:从SD大鼠皮下脂肪获取的脂肪源性内皮细胞经过成脂诱导后可转化为脂肪细胞,这对研究脂肪细胞的来源和形成以及脂肪组织工程研究具有重要的意义。
Objective:To study the adipogenous potential of endothelial cells derived from SD rat adipose tissue. Methods:The subcutaneous adipose tissue was obtained from the fold inguen of four SD rats, then digested with collagenase type Ⅰ and cultured in endothelial medium (Gibco) for seven days. When the cells appeared endothelial characteristics identified by transmission electron microscopy and immunocytochemical staining with factor anti wm antibody, the cellss were cultured in adipogenic inducing medium (supplemented with insulin, 1-methyl-3-isobutylxanthine, indomethine and dexamethasone) for seven days and stained with Oil Red O. Results: The adipose tissue derived endothelial cells uniformly had characteristic cobblestone morphology and arrangement. After 7-10 d culture in endothelial medium,95% of the cells were factor vm positive and Weibel-Palade body was found in cytoplasm of some cells. After seven days of adipogenic induction, about 85% of the cells displayed accumulation of lipid vacuoles detected by Red Oil O staining. Conclusion: Endothelial cells from adipose tissue can be induced to differentiate into adipose cells.
