摘要
中国健康志愿者10名,一次口服硫酸吗啡普通片20mg,分别于服药后不同时间采取静脉血,用GC-MS测定血浆中吗啡含量。以3P87软件程序处理,求得其主要药代动力学参数:Cmax为21.27±6.21μg/L;Tmax为0.55±0.16h;曲线下面积AUC为93.08±16.65μg·h-1·L-1,T1/2β为3.03±0.74h。结果提示,硫酸吗啡普通片在人体内的吸收,消除均较快,并表明吗啡的最大血药浓度与临床的最强镇痛作用之间存在一个迟滞时间。
The pharmacokinetics of morphine sulphate was studied in 10 Chinese healthy volunteers after a single oral dose(20mg).Blood samples were collected at 0.25,0.5,1,1 .5,2,3,4,6,8 and 10 h after administration,and determined by GC MS method for morphine concentration in plasma. The pharmacokinetic parameters calculated according to 3P87 program were as following:Tmax=0.56±0.16h Cmax=21.27±6.21μg/L ,CL=220.4±34.7 L/h,T1/2β=3.03±0.74 h.The results showed that morphine was characterized by a rapid absorption and elimination. There was a lag time between the maximum analgesic effect and maximum plasrna concentration.
