缺氧性血管收缩与内皮依赖性NO缺陷相关性实验研究

摘要 目的探讨缺氧性血管收缩对Wistar大鼠NO活性的影响。方法制备Wistar大鼠内皮完整和去内皮的胸主动脉环,缺氧刺激动脉环,记录收缩幅度的变化。结果内皮细胞完整、去内皮血管环缺氧时收缩幅度分别为598±42mg、208±38mg,两者比较有显著性差异(P<0.01);内皮细胞完整血管环在缺氧同时分别应用L-NNA、L-Arg、NTG和MB,可分别使血管收缩增幅达64.88%(P<0.01)、降幅达25.08%(P<0.05)、降幅达45.15%(P<0.01)和增幅达42.07%(P<0.01);去内皮血管条在缺氧时应用L-Arg或L-NNA,并不明显改善血管收缩幅度(P>0.05),而NTG使血管收缩降幅度达57.21%(P<0·01),MB部分抑制NTG的作用,血管收缩增福88.76%(P<0.01)。结论上述结果提示,内皮细胞参与缺氧性血管收缩反应,缺氧时内皮细胞依赖性合成和释放NO不足,可能是血管收缩反应的机制之一。 Objective To observe effect of large blood vessel hypoxia on the Wister rat's NO activity. Methods Thoracic aorta rings model with full endothelium and removed endothelium of Wistar rat were prepared , hypoxia stimulated aorta rings and noted changes of constriction extent. Results Endothelium full , removed endothelium aorta tings by hypoxia contract extent difference was 598 ±42mg and 208 ± 38mg, both had significance discrepancy (P 〈 0.01 ) fairly; The endothelium full aorta rings by hypoxia at the same time applied L - NNA, L - Arg, NTG and MB respectively, can make blood vessel contract to increase respectively reach 64.88% ( P 〈 0.01 ) ,reduce reach 25.08% ( P 〈 0.05 ) , reduce reach 45.15 % ( P 〈 0.01 ) and increase reach 42.07% ( P 〈 0.01 ) ;The removed endothe lium aorta rings by hypoxia application L - Arg or L - NNA, and did not improve blood vessel obviously to contract range (P 〉 0.05 ) ,and NTG made blood vessel contract to reduce range, reach 57.21% ( P 〈 0.01 ) , MB inhibited the role of NTG partially, blood vessel contracted to increase the 88.76% (P 〈 0.01 ). Conclusions These results susgest that endothelial cell participates in anoxic blood vessel constraction , in the hypoxia synthesization and release of endothelial cell del?endence NO is deficiency , it maybe one of mechanisms of blood vessel constraction.
