摘要
目的:观察中药天年饮对衰老大鼠睾丸形态结构的影响。方法:实验于2004-04/07在承德医学院基础医学研究所完成。①选用清洁级4月龄雄性SD大鼠40只,随机分为4组:正常组、衰老模型组、天年饮用药组、阴性对照组,10只/组。天年饮由何首乌、怀牛膝、肉苁蓉、淫羊藿、丹参、茯苓6味中药组成,经熬制后每mL含生药1.5g。②衰老模型组、天年饮用药组、阴性对照组大鼠均采用D-半乳糖连续腹腔注射建立亚急性衰老大鼠模型。造模完毕后,天年饮用药组每天按8.1g/kg灌胃给予天年饮,共30d;阴性对照组每天灌胃同等剂量的生理盐水,共30d;正常组、衰老模型组大鼠不再作任何处理。③采用ELISA方法检测各组大鼠血清睾酮含量,苏木精-伊红染色观察睾丸生精小管的形态结构。结果:实验选用雄性SD大鼠40只,全部进入结果分析。①各组大鼠血清睾酮含量的检测结果:与正常组大鼠比较,衰老模型组、阴性对照组大鼠血清睾酮含量均明显降低眼(1.26±0.32),(0.52±0.14),(0.59±0.17)μg/L,P<0.05演;天年饮用药组大鼠血清睾酮含量与模型组大鼠相比明显升高眼(1.16±0.32),(0.59±0.17)μg/L,P<0.05演。②大鼠睾丸生精小管形态结构的变化:天年饮用药组大鼠睾丸生精小管的形态结构较衰老模型组明显恢复,基膜趋于完整,生精细胞层数增多。但与正常组大鼠睾丸相比,生精细胞仍显较稀疏。结论:中药天年饮可明显抑制衰老大鼠血清睾酮含量的下降,改善睾丸的形态结构,具有一定延缓衰老的作用。
AIM: To observe the effects of traditional Chinese medicine Tiannianyin on testis morphology of aging rats. METHODS: The experiment was carried out in the Institute of Basic Medical Sciences of Chengde Medical College from April to July 2004. ① Totally 40 male SD rats aged 4 months were applied in this study and divided into 4 groups at random: normal group, aging model group, Tiannianyin group, and negative control group with 10 rats in each group. Tiannianyin was consisted of tuber fleeceflower root, achyranthes root, desertliving cistanche, epimedium, danshen and tuckahoe, and had 1.5 g crude drug in every mL. ②The subacutely aging model rats were made by injecting D-gal into abdominal cavity continually in model group, Tianni-anyin group and negative control group rats. After the models were successfully established, no treatment was given to the rats in normal group and model group; the rats in Tiannianyin group were treated with gastric perfusion of 8.1 g/kg Tiannianyin, once a day for 30 days, and those in negative control group were intragastrically perfused with saline of the same volume and same time for 30 days. ③ELISA was used to detect the testosterone content in serum, hematoxylin-eosin staining (HE) to observe the morphology of seminiferous tubule. RESULTS: All the 40 rats were involved in the analysis of results. ①The results of testosterone content: Compared with normal rats, the testosterone content of rats in aging model group and negative control group decreased significantly [(1.26±0.32), (0.52±0.14), (0.59±0.17)μg/L,P 〈 0.05 ). The testosterone content of rats in Tiannianyin group increased obviously as compared with aging model rats [(1.16±0.32), (0.59±0.17) μg/L,P 〈 0.05), ②Changes of morphology of seminiferous tubule of rats: The morphology of seminiferous tubule in Tiannianyin group obviously recovered as compared with aging model group; The basal lamina was mostly integral and layer number of spermatogenic cells increased, but
