摘要
目的研究糖维康(TWK)对胰岛素抵抗(IR)大鼠血糖、胰岛素、瘦素(Leptin)、肿瘤坏死因子(TNF-α)及C-肽(C-P)水平的影响,探讨其保护IR大鼠的机制.方法采用高脂高糖饲料喂养法复制IR大鼠动物模型,同时给予不同剂量的TWK干预,实验10wk后采用葡萄糖氧化酶法测定血糖,采用放射免疫分析方法检测血中胰岛素、瘦素、TNF-α及C-肽水平.结果TWK生药1.5g/mL,生药0.75g/mL能够增加IR大鼠的胰岛素敏感指数(ISI)(P<0.01),降低其血清瘦素水平(P<0.05);TWK生药1.5g/mL与二甲双胍(Met)104mg/kg合用可降低IR大鼠血清瘦素、TNF-α水平(P<0.01),升高C-肽水平(P<0.05),TWK生药0.75g/mL升高C-肽水平(P<0.05).结论TWK可通过降低血清瘦素水平,协同Met降低血清TNF-α水平,增加胰岛B细胞功能,改善IR大鼠的高胰岛素状态.
AIM: To study the effect of Tangweikang on blood glucose, serum insulin, leptin, TNF-α and C-peptide in rats with insulin resistance (IR). METHODS: The IR rats were induced by administration of high-fat and high-sugar diet. At the same time, the different dose of Tangweikang was given to the rats. After 10 weeks, the blood glucose was tested by the glucose oxidase method and serum insulin, leptin, TNF-α and C-peptide were determined by radioimmunoassay in the rats. RESULTS: Tangweikang 1.5 g/mL, 0.75 g/mL increased the IR rats' insulin sensitivity index (ISI) ( P 〈 0, 01 ), decreased serum leptin (P 〈 0, 05 ). Combining Tangweikang 1.5 g/mL and Metformin 104 mg/kg decreased the levels of serum leptin and TNF-α (P 〈 0. 01 ), and increased C-peptide ( P 〈 0. 05 ) in IR rats, Tangweikang 0, 75 g/mL increased IR rats' C-peptide level (P 〈 0.05). CONCLUSION: Tangweikang can improve ISI of IR rats, by decreasing serum leptin, enhancing Met's effect to reduce the level of serum TNF-α and increasing C-peptide,
