摘要
目的研究肝细胞癌(HCC)病人、乙型肝炎病毒(HBV)携带者、健康人血清蛋白质表达的差异,以建立HCC血清蛋白质指纹图谱筛选模型。方法用表面加强激光解析电离飞行时间质谱技术(SELDI-TOF-MS)获得HCC病人、HBV携带者及健康人血清中的蛋白质指纹图谱,用计算机软件比较分析,并建立HCC的血清蛋白质指纹图谱模型筛选模型。结果HCC组与非肿瘤组共有11种蛋白质有显著性差异;以其中4种蛋白质生物标志物(相对分子质量3 448、6 8834、467和3 134)组建的筛选模型检测灵敏度为94.6%,特异度为92.3%。结论建立的血清蛋白质指纹图谱模型能够区分HBV携带者与HCC病人,SELDI-TOF-MS在HCC的诊断及肿瘤特异性蛋白质生物标志分子的筛选等方面具有一定价值。
Objective To establish a serum proteomic pattern and study the differences of serum protein spectra of patients with hepatocellular carcinoma (HCC), of HBV carriers and of healthy controls. Methods Proteomic spectra of HCC and age and sex-matched non-HCC controls, including HBsAg carriers and healthy peoples, were generated by surface-enhanced laser desorption/ionization time of-flight mass spectrometry (SELDI TOF-MS) on WCX-2 chips. Ciphergen software was used to identify proteomic features, and Biomaker Wizard software to detect the differences in protein peaks between HCC and non-HCC controis, and then to set up a serum biomarker pattern to screen the HCC. Results Eleven proteins were significantly different be tween HCC and non HCC controls, among which, four (m/z 3 448.6 883.4 467 and 3 134) were selected to set up a optimal serum biomarker pattern. This proteomic pattern yielded a high sensitivity (94.6%) and specificity (92.3%). Conclusion The estab lished serum proteomic pattern can distinguish the HCC cases from non HCC ones, and the proteomics approach of SELDI-TOFMS can greatly facilitate the discovery of new tumor biomarkers.
