摘要
目的:观察HL-60细胞株经顺铂作用和60Co照射后其细胞周期阻滞的动态变化。方法:用60Co分别以6,10Gy的吸收剂量照射HL-60肿瘤细胞株,并于照射后6、12、24及60h测量其细胞周期变化,同样使用化疗药物顺铂分别以10μmol/L,20μmol/L浓度梯度作用HL-60细胞株,并于作用后4、8、12及24h测量其细胞周期变化。结果:HL-60细胞株在受到小剂量外界作用后,如6Gy60Co照射和10μmol/L顺铂作用,表现有G2/M期崩溃现象,而在大剂量外界作用后,如10Gy60Co照射和20μmol/L顺铂作用,未表现G2/M期崩溃现象,而是表现持续的细胞周期阻滞现象。结论:在一定范围的外界剂量作用下,HL-60细胞株的细胞周期阻滞现象即自我保护机制呈剂量依赖性增强。小剂量外界刺激可导致其凋亡增加,而大剂量反而使其表现较强的细胞周期阻滞现象。
Objective:To observe the cell cycle arrest changes of the HL-60 leukemic cell line after ^60Co irradiation and cisplatin treatment. Method: HL-60 tumor cell line was irradiated with ^60 Co at the absorbed doses of 6, 10 Gy. Then the cell cycles change was checked at the time of 6, 12, 24 and 60 h after irradiation. HL60 cell line was treated with cisplatin at the concentration gradient of 10 μmol/L and 20 μmol/L, Then the cell cycles change was checked at the time of 4, 8, 12 and 24 h after treatment. Result: HL-60 cell line manifested G2/M phase collapse phenomenon and apoptosis increased after the low dosage treatment such as 6 Gy ^60 Co irradiation and 10 μol/L cisplatin. But it didn't manifested G2/M phase collapse phenomenon at the higher dosage treatment such as 10 Gy 60 Co irradiation and 20 μmol/L cisplatin, it manifested consistent cell cycle arrest. Conclusion:Within a certain dosage range, the HL-60 cell cycle arrest increased in a dose-dependent manner, that is, apoptosis increased after the low dosage treatment, but it manifested stronger cell cvcle arrest after the higer dosage treatment.
