摘要
目的探讨儿童慢性乙型肝炎(CHB)和儿童乙型肝炎病毒(HBV)携带者(ASCs)的致病机制。方法采用抑制消减杂交(SSH)技术构建两者外周血单个核细胞差异表达基因cDNA文库,得到差异表达基因β2微球蛋白(β2-MG)。采集34例CHB和41例ASCs儿童外周血血浆,用酶联免疫试验检测β2-MG水平。结果成功构建了儿童CHB和ASCs的差异表达文库,发现在CHB儿童中β2-MG和铁转运蛋白等炎症相关基因表达增高。证实了β2-MG在蛋白水平也存在相应的差异表达。结论β2-MG在CHB患儿中的高表达参与儿童CHB的发病。
Objective To explore the difference of pathogenesis between chronic hepatitis B (CHB) and asymptomatic hepatitis B virus(HBV) carriers(ASCs) in children. Methods Subtracted libraries was constructed by suppression subtractive hybridization (SSH) from peripheral blood mononuelear cell (PBMC) of two groups, beta- 2 - mieroglobulin (β2 - MG) was screened as one of differentially expressed genes. Serum was collected from children with CHB and ASCs, the differential expression of β2 - MG was confirmed by enzyme imm^y, Results Subtractive libraries were constructed successfully, the differentially expression of β2 - MG and lactroferrin were up - regulated in CHB children. And the differentially expression of β2 - MG on the level of protein was confirmed. Condusion The upregulation of β2 - MG in CHB children is involved in the pathogenesis mechanisms.
