摘要
目的观察乌头注射液抗炎作用的强弱及其药效动力学过程,求出相应的药效动力学参数,为合理用药提供参考。方法分别用巴豆油和琼脂所致的小鼠耳肿模型和足肿模型,以耳片重量和足肿体积为指标研究了乌头注射液的抗炎作用,以与空白对照组相比,一次给药后足肿程度变化的经时过程进行房室模型拟合和药效动力学参数估算。结果显示乌头注射液具有显著的抗炎作用,其药效消除呈一级动力学过程并基本符合血管外给药的一房室开放模型,其上升相的速率常数Ka=1.876 1 h-1,药效倍增期为0.37 h;其消除相的速率常数K=0.136 2 h-1,半衰期t1/2E=5.09 h;其达峰时间为2.21 h;持续时间约为8.67 h。结论乌头注射液具有显著的抗炎作用,求算其药效动力学参数,可为合理用药作参考。
OBJECTIVE To study the anti-inflannuation effects and pharmacodynamice of Wuto injection. METHODS The model of swelling ear and back -metatrarsal in mice was built and the swelling extent was measured to study the anti-inflammation. The apparent parameters of pharmacodynatnics were estimated based on the time-effect curve. RESULTS Wutou injection has anti-inflammatory effect obviously, Thy effect-time curve was described by an one-compartment model of extravascular administration. The pharmacodynamic parameters were: Ka=1.8761h^-1, t1/2a=0.37h, K=0.1362h^-1, t1/2F=5.09h, tmax=2.21h, respectively. CONCLUSION Wutou injection has anti-inflammatory effect obviously. The pharmacodynamic parameters may be helpful for clinical practice.
