NBS法标记CEA人鼠嵌合抗体rch24的初步研究

摘要 目的:研究125I直接标记CEA人鼠嵌合抗体rch24的方法。方法:采用NBS(溴代琥珀酰亚胺)法进行碘标,并测定标记物的放射化学纯度、标记率和体外稳定性。结果:采用纸层析法测定的标记率和放射化学纯度分别为97.2%±0.4%和95.5%±0.2%;标记抗体体外4℃存放,放射化学纯度逐渐下降,14天后降为92.6%,35天后为89.3%。结论:标记物具有较高的放射化学纯度及稳定性,为体内治疗打下良好的基础。 Objective: To study the characteristics of ^125I radiolabeled anti-CEA human-mouse chimeric antibodies rch24. Methods: Iodine labeling with ^125I was performed with NBS method, The labeling efficiency, radiochemical purity and stability in vitro were determined. Results: The labeling efficiency was 97.2%± 0.4%, and the radiochemical purity was 95.5% ±0.2%. The radiochemical purity was decreased to 92.6% and 89.3% when the labeled antibodies were stored at 4℃ for 14 d or 35 d. Conclusion: The radiolabeled antibodies have high radiochemical purity and stability which lays basis for therapy in vivo.
