摘要
利用ABC免疫转印技术分析一组共16个抗大肠癌McAbs识别抗原的特性,效果满意。该法不需分离纯化抗原和制备腹水,使用杂交瘤培养上清液即可得到清晰,准确的识别抗原区带,背景浅淡,用来分析识别抗原分子量及理化性质,特异性强、重复性好、敏感度高。可检测的最小肿瘤抗原蛋白量为1.25×10^(-6)mg,明显优于PAP免疫转印和间接酶免疫转印法。本文提示,ABC免疫转印是分析复杂而微量的抗原的有效方法。
Characters of 36 identified antigens of anti-colon cancer McAb were analysed by immunoblot technique with avidin-biotin peroxidase complex (ABC-Ib). The results showed that ABC-Ib has high sensitivity, reproducibility and specificity. Clear and accurate identified antigen band on a light background was gained with only culture supernatant of hybrid. The minimum amount of protein of tumer tissue antigen that can be detected is 1.25 × 10- 6mg. It is suggested that ABC-Ib is very effective in studying the complex and small amount antigen.
