摘要
目的建立补骨脂注射液中补骨脂素和异补骨脂素高效液相含量测定方法,并探讨补骨脂素和异补骨脂素浓度变化的可能因素。方法用Nucleosil C18色谱柱,柱温35℃;检测波长247nm;流动相:甲醇-0.1%冰醋酸(55:45)。结果线性范围分别为:补骨脂素:1.1~11μg,r=0.9990;异补骨脂素:2.6~26μg,r=0.9991。平均回收率为98.01%(RDS=3.37%)、96.90(RDS=2.44%)。100 ℃加热5h补骨脂素、异补骨脂素含量无差异。改变pH值条件加热处理,补骨脂素、异补骨脂素浓度比有改变。结论本方法可用于补骨脂注射液中补骨脂素、异补骨脂素的含量测定。pH值的改变,可导致补骨脂素和异补骨脂素的转化或降解。
OBJECTIVE To establish an HPLC method for assay of psoralen and isopsorale in Fructus Psoraleae injection . To observe the factors which the content change. METHODS Separation was performed on a Nucleosil C18 column, column tempreture was 35℃. The UV detect wavelength was set at 247nm. The mobile phase consisted of methanol-0.1% acetic acid ( 55: 45 ). RESULTS The linear range of psoralen was 1.1 μg - 11 μg ( r = 0.999 0), of isopsorale was 2.6μg - 26μg ( r = 0. 999 1 ). The average recoveries were 98.01% ( RDS = 3.37 % ) and 96.90 ( RDS = 2.44% ), After being heated at 100℃ for 5 hours, the content mesurement of them were not different from before. But it were different with the change of pH. CONCLUSION The method could be applied to determinate the content of psoralen and isopsorale in Fructus Psoraleae injection. The result showed that psoralen coulden' t be transformed into isopsorale only by heating. The transformation or decomposition would be made by the change of pH.
