摘要 目的 建立人血浆中普卢利沙星活性代谢产物NM394的高效液相色谱测定方法。方法 取血浆样品0.5mL,用高氯酸沉淀蛋白,以乙腈-枸橼酸溶液为流动相,用Hyperisl BDS2柱分离,紫外检测器272nm处检测。结果 普卢利沙星活性代谢产物NM394的线性范围为0.025~5.0μg·mL^-1,平均回收率为107.0%~114.0%,日内、日间精密度均〈10%。结论 方法灵敏、专属、快速,可用于普卢利沙星人体药动学及生物利用度研究。 OBJECTIVE To establish a method to determine NM394 concentration in the plasma by HPLC.METHODS The plasma 0. 5 mL was depurated with perchloric acid, and then determined by HPLC with Hyperisl BDS2 column, acetonitrile-citric acid solution as the mobile phase and the detection wavelength was 266 nm. RESULTS The calibration curve was linear within 0. 025~5.0μg·mL^-1; the recovery was 107.0%~114.0% ; RSD of intraday and inter-day was no more than 10%. CONCLUSIONS The method is accurate and sensitive with no endogenous interference, and is suitable to investigate the pharmacokinetics and relative bioavailability of prulifloxacin.
