摘要
目的分析结核分枝杆菌全细胞脂肪酸的种类与数量构成,以进一步揭示其生物学特征,为结核分枝杆菌的其它基础研究提供依据。方法应用Middlebrook 7H10琼脂培养基对临床分离培养的初代分枝杆菌培养物进行纯培养,对被鉴定为结核分枝杆菌的87株及结核分枝杆菌标准株H37,Rv、H37,Ra各1株应用气相色谱技术、采用蜂面积归一化法定量分析碱处理细菌细胞释放出来并甲基化的细胞脂肪酸。结果1.各菌株均含有C14:0、C16:0、C18:0、C18,1ω9、10-Me-C18:0脂肪酸,其平均相对含量分别为:2.08%、39.78%、10.09%、21.21%和14.17%。C15:0、C18,1ω6、C17:0、C20:0脂肪酸为90%以上菌株含有组分,其平均相对含量分别为:1.15%、4.07%、2.91%和1.38%。C12.0、C16.1ω9、C18,1ω7、8-Me-C18:0、10-Me-C18:0、C17,1ω7、C17,1ω8为含量少且为只有部分菌株具有的细胞脂肪酸组分。所有菌株均不含C9、C10、C11、C13、C19脂肪酸。2.临床分离株脂肪酸的相对含量构成与H37Rv接近,而与H37R有较大差别。结论在C9~C20脂肪酸中,结核分枝杆菌主要含有C16:0、10-Me-C18:0、C18:0饱和脂肪酸与C18,1ω9、C16,1ω不饱和脂肪酸,并提示脂肪酸的组成特征可能与结核分枝杆菌毒力有关。
To determine the kinds and quantities of the whole cell C9-C20 fatty acid ingredients from Mycobacterium tuberculosis for further revealing its biological characteristic and providing more information for other basic researches, cultures of the primary clinical isolates were purified by using Middlebrook 7H10 agar culture medium into pure culture strain, and 87 strans of clinical isolates and each of one strain of the standard stains H37Rv and H37Ra were subjected to be analyzed by means of gas chromatography and peak area normalization method to detect the presence of fatty acids liberated from bacterial cells in condition of strong alkaline and methylated subsequently. It was demonstrated that all strains investigated contained C14:0C16:0C18:0, C18:1ω9, and 10-MeC18:0 fatty acids with their relative average contents of 2.08%, 39.73%, 21.23%, 10. 13% and 14.20% respectively, and C15:0, C18 ,1ω 6, C17:0. C20:0 fatty acids existed in 90% of strains with their relative average contents of 1.15%, 4.07%, 2.91% and 1.38% respectively. Other strains with 1ow-content of fatty acids or with fatty acids contained in part strains only included C12:0. C16:1ω9. C16:1ω7. 8-Me-C16:0 10-Me-C16:0, C17:1ω7. C17:1 ω8. fatty acid . No C9. C10, C11. C13 and C19, fatty acids could be detected in all strains. The relative average contents of fatty acids in the clinical isolates approached to that of the standard strain H37Rv, but showed great differences with that of the strain H37Ra. It is evident that , among the C9-C20 fatty acids, M. tuberculosis mainly contains saturated fatty acids C16:0, 10-Me-C16:0 and C18:0 as well as unsaturated fatty acids C18 :1ω9 and C16:1ω ,suggesting that composition pattern of the fatty acids from M. tuberculosis might be associated with its virulence.
